#5 Homo Demens
Homo Demens is introduced as a major enemy faction as a Militant separatist group in the game whose leader is Higgs, aka The Man in the Golden Mask revealed in the earlier trailer, the character played by the famous voice actor Troy Baker. The trailer also indicates that the game will feature more than one enemy factions/terrorists group. Whatsoever be the case, it seems Higgs (Troy Baker's character) seems to be far more important than the rest of them.
#6 Something called "Edge Knot City"
The trailer reveals that Edge Knot City is one of the major cities in this crumbling divided world of Death Stranding and that these Homo Demens reside there or are at least fighting for its independence. Does this mean other cities are overrun by other enemy factions? Well, it seems more likely that is the case.
#7 Madds Mikkelsen's character
The character which had the most screen time in this latest trailer aside Sam is Madds Mikkelsen's character called Cliff. He appears thrice in the trailer. Once in the beginning wearing suits and talking to a "baby". The next time he appears is in the game's so called "Hades" where the player ventures into once they die.
Hades seems to repesent a part of the past hence that's the reason it's set amidst the backdrop of a war setting. It seems though Madds character Cliff is somehow stuck in there as the trailer indicates in quotes,

#8 The connection between Higgs and Cliff's character
One thing we surely know is that Higgs and Cliff both serves as a threat to Sam as well as the shattered nation he's trying to reconnect. At the end of the trailer, we hear Higgs speak that he's fortunate of having a good connection to the other side which is Hades, the place where Cliff is stuck. And the final shot of a jungle where Cliff is seen with his skeleton soldiers, it seems somehow he made his way back into the real world, probably with the help of Higgs?
#9 Multiplayer component
Death Stranding will feature multiplayer components but not in a traditional way you would expect. Apparently the game will feature asynchronous online gameplay where a player can send supplies, share safe houses and walk in the footsteps of fellow couriers to reunite civilisation.
#10 Overall plot
The summary of plot listed on the PS Store preorder page states the following,
Sam Bridges must brave a world utterly transformed by the Death Stranding. Carrying the stranded remnants of the future in his hands, Sam embarks on a journey to reunite the shattered world one step at a time.
