The Sabres signed Nylander shortly after the draft, and he spent most of his first season with their American Hockey League (AHL) affiliate, the Rochester Americans.

Nylander made his NHL debut on April 3, 2017, against none other than the Toronto Maple Leafs, and scored his first NHL goal just a few days later against the Tampa Bay Lightning. After scoring 12 goals for Rochester in the 2018-19 season, Nylander was given a chance with the Sabres and scored twice in 12 games.

In July 2019, Nylander was traded to the Chicago Blackhawks, who gave him his father's old jersey number, 92, and a regular role in the lineup during the 2019-20 season. Nylander scored 10 goals and 26 points in 65 games, showing flashes of the offensive potential that made him a high draft pick.
