In the Hotel Fitness Championship, he finished eighth, second in the Small Business Connection Championship, and first in the Web.com Tour Championship.

Grillo secured his eighth career PGA Tour victory and his first as a member by winning the 2016 Frys.com Open, the season-opening event of the PGA Tour, after a sudden-death playoff against Kevin Na. Grillo scored a 3-under-par 69 and holed a 25-foot birdie putt on the 72nd hole, which Na later matched. Although Grillo missed a three-foot putt for victory on the first playoff hole, he won with a birdie on the second hole as Na could only manage a par after hitting a shot behind a tree.
