She attended the University of Connecticut in 2006 and began playing for the UConn Huskies women's basketball team. Over her college career, she played in 152 games, scoring 2,346 points and grabbing 1,367 rebounds, making her the all-time leading scorer and rebounder at the University of Connecticut. She won two consecutive national championships in 2009 and 2010.

Charles was selected by the Connecticut Sun as the first overall pick in the 2010 WNBA draft. She played four seasons for the Sun before being traded to the New York Liberty in the 2014 WNBA draft. She has also played for the Washington Mystics, the Phoenix Mercury, and the Seattle Storm.
