She attended the University of Connecticut from 2011 to 2015, where she played for the UConn Huskies women's basketball team. Stokes played in 146 games over her four-year college career, recording 602 points and 819 rebounds. She helped the Huskies win three national championships from 2013 to 2015. She was also named the 2014-15 American Athletic Conference Defensive Player of the Year and was selected for the 2014-15 All-Athletic Conference Second Team.

Stokes made her WNBA debut in 2015 for the New York Liberty as the 11th overall pick. She played six seasons for the New York Liberty, from the 2015 to 2021 WNBA seasons, before moving to the Las Vegas Aces during the 2021 WNBA season.
