Jefferson grew up in Navarre, Florida, and attended Navarre High School, where he participated in football and wrestling.He was awarded first-team all-area after totaling 60 tackles, 22 tackles-for-loss (TFLs), and five sacks as a senior in 2018; he was also named to the Florida Athletic Coaches Association All-Star Game and finished his career at Navarre with 115 tackles, 35 TFLs, and 11 sacks. He committed to play collegiate football for the West Virginia Mountaineers after receiving a three-star recruit ranking.

Jefferson played 11 games as a true freshman at West Virginia in 2019, including one as a starter, and recorded five tackles. His playing time decreased, as he made only one tackle in the 2020 season. He had more playing time in 2021, recording 17 tackles and four TFLs. The Athletic named Jefferson to their "Freak List" for the 2022 season. That season, he was a full-time starter with 31 tackles, 9.5 TFLs, five pass breakups, and three sacks. Jefferson entered the NCAA transfer portal after the 2022 season. He eventually transferred to the LSU Tigers. In his one season with the Tigers, he recorded 36 tackles, seven TFLs, and 2.5 sacks. He was invited to attend the 2024 Senior Bowl and NFL Scouting Combine.
