Mobley played college basketball for the USC Trojans, which represented the University of Southern California. He appeared in 33 games in his freshman year, scoring 540 points and 286 rebounds as a forward.

Before his sophomore year, Mobley declared for the 2021 NBA draft and was one of the top prospects for the draft. He was drafted as the third pick by the Cleveland Cavaliers in the first round of the draft that year. Mobley debuted in the NBA regular season against the Memphis Grizzlies; he posted 17 points and nine rebounds, along with six assists.
