In his debut game in the 2017 season against the Denver Broncos Ekeler recorded two receptions for 18 yards. He scored the first touchdown of his NFL career in a game against Philadelphia Eagles. Ekeler continued his brilliant performance in the upcoming seasons for the Chargers where in the 2021 season he recorded 20 touchdowns in the entire season and became the only second person in NFL history to do so. Ekeler was ranked by his team members at 46th in the list of Top players of NFL 2022. Ekeler had another great season in 2022 where in a match against the Los Angeles Rams he recorded 122 yards and had 39 receiving yards and two touchdowns and due to his brilliant performance he was named as the AFC Offensive Player of the Week.

After playing on a one-year deal with the Chargers in 2023, Ekeler signed a deal with the Washington Commanders in March 2024.
