The Minnesota Timberwolves didn't hesitate and proudly selected him. By July, Towns had signed his rookie scale contract, and by October, he was making waves on the court, debuting against the Los Angeles Lakers. His early promise was further solidified in the subsequent game against the Denver Nuggets. With an astonishing 28 points and 14 rebounds, Towns was instrumental in securing the Timberwolves their historic first-ever 2-0 start with both victories on the road.

2016 solidified Towns' position among the elite. At just 21, he became the third-youngest player in three decades to record at least 45 points and 15 rebounds in a game. Furthermore, he reached his 100th career double-double, standing behind only Dwight Howard as the second-youngest player in NBA history to achieve this milestone. In 2022, Karl-Anthony Towns had to rest in the third quarter due to his non-contact calf injury. According to the sources, the injury was a Grade 2 injury, so he had to miss 6 weeks from the game. But in January 2023, Towns stated he had a Grade 3 injury which would take around 2 months to get back. In April Towns was back to the court and played the final game against the New Orleans Pelicans.
